Interpolated concentrations of native Tissue Factor in mouse brain cell culture supernatant and mouse urine samples.
The concentrations of Tissue Factor were measured in duplicates, interpolated from the Tissue Factor standard curves and corrected for sample dilution. Undiluted samples are as follows : mouse brain cell culture supernatant 100%, mouse urine 100%. For mouse urine, dilutions beyond 2 fold were not analysed due to O.D. values below the O.D. values of the MDD. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Tissue Factor concentration was determined to be 79 pg/mL in mouse brain cell culture supernatant and 11 pg/mL in mouse urine.

The concentrations of Tissue Factor were measured in duplicate and interpolated from the Tissue Factor standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Tissue Factor concentration was determined to be 713 pg/mL in Mouse Brain Tissue Extract, 206 pg/mL in mouse C2C12 Cell Extract and 824 pg/mL in NIH/3T3 Cell Extract.

Mouse Tissue Factor ELISA Kit (ab214091) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Tissue Factor protein in cell culture extracts, cell culture supernatant, tissue extracts, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Tissue Factor with 3.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Tissue factor plays a role in normal hemostasis by initiating the cell-surface assembly and propagation of the coagulation protease cascade. Tissue Factor initiates blood coagulation by forming a complex with circulating factor VII or VIIa. The tissue factor-VIIa complex activates factors IX or X by specific limited protolysis.

Biological function summary
Tissue factor acts as a cofactor for factor VIIa forming a complex that rapidly catalyzes the activation of factor X to factor Xa. This catalytic action ultimately leads to fibrin formation which is essential for clotting. Tissue Factor (TF) is part of the extrinsic pathway of the coagulation cascade and is fundamental in the generation of a hemostatic response to tissue injury. By regulating thrombin generation through its interaction with other coagulation factors tissue factor plays an important role in maintaining hemostatic balance.
Pathways
Tissue factor is integral in the extrinsic pathway of coagulation which works closely with the intrinsic pathway to ensure effective hemostasis. It complexes with factor VIIa and triggers downstream effects involving proteins like factor X and thrombin. Tissue factor forms a bridge between cellular injury responses and the blood coagulation system linking these two significant physiological processes. The interaction with these proteins highlights its central role in hemostatic pathways and highlights its significance in the coagulation network.
