Interpolated concentrations of native TLR2 in Mouse cell and tissue extract samples.
The concentrations of TLR2 were measured in duplicate and interpolated from the TLR2 standard curve and corrected for sample dilution and extract load. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TLR2 concentration was determined to be 11.1 pg/μg in J774A.1 Cell Extract and 1.75 pg/μg in Spleen Tissue Extract.

Mouse TLR2 ELISA Kit (ab224880) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TLR2 protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TLR2 with 88 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less

Toll-Like Receptor 2 (TLR2) is a transmembrane receptor, expressed on a wide variety of cell types including microglia, monocytes, macrophages, and dendritic cells. TLR2 cooperates with multiple other proteins (TLR1, LY96, TLR6, and other) to mediate an immune response to bacterial lipoproteins. Cytokine secretion in response to these bacterium is found to occur in a TLR2 dependent fashion. ab224880 recognizes the extracellular domain of the transmembrane receptor.

Biological function summary
TLR2 initiates signaling pathways that lead to the production of cytokines and chemokines helping to control infection. It forms dimers with other TLRs like TLR1 or TLR6 to expand its recognition range of microbial components. TLR2's activity is important for balancing pathogen recognition and avoiding overactive immune response. On macrophages TLR2 activation enhances their phagocytic ability.
Pathways
TLR2 plays roles in innate immune signaling pathways and inflammation pathways. It contributes significantly to the NF-kB and MAPK pathways which regulate inflammation and immune response. TLR2 often cooperates with proteins like MyD88 and TRAF6 in these pathways leading to downstream activation of transcription factors and production of pro-inflammatory molecules.
