Abcam's mouse TrkB in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Mouse TrkB in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
A TrkB specific rat monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat specific for TrkB is then added followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the mouse TrkB amount of sample captured in plate.

Biological function summary
TrkB plays essential roles in neuronal survival differentiation and synaptic plasticity. It acts as an important mediator for the effects of neurotrophins primarily BDNF on neurons. While TrkB itself is not part of a multi-protein complex it interacts closely with several adaptor proteins post-activation to propagate survival and proliferation signals within the neuron. These biochemical interactions greatly influence neurodevelopment and cognitive functions.
Pathways
TrkB is a pivotal player in both the MAPK/ERK and PI3K/AKT pathways. Upon BDNF binding TrkB undergoes autophosphorylation triggering these pathways which are critically involved in cell survival and growth. Other proteins such as SHC1 and PLCγ interact with TrkB in these pathways extending the range of cellular responses to neurotrophin signaling facilitating processes like neuronal plasticity and memory formation.
