Product details

Abcam's Tweak Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse Tweak in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Tweak coated on a 96-well plate. Standards and samples are pipetted into the wells and Tweak present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse Tweak antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Tweak bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

TWEAK also known as Tumor Necrosis Factor-like Weak Inducer of Apoptosis is a type II transmembrane cytokine with a mass of approximately 24 kDa. This protein identified by the human gene TNFSF12 is widely expressed in several tissues including the heart pancreas and skeletal muscle. TWEAK also exists in a soluble form which can arise through proteolytic cleavage. The protein plays various roles in cellular functions by interacting with its receptor Fn14 initiating signaling cascades that affect cell fate.
Biological function summary

TWEAK influences processes such as cell proliferation differentiation and apoptosis. It does not form part of a larger complex but functions independently to mediate cellular activities. The interaction with Fn14 its sole receptor leads to the activation of several downstream signaling pathways. TWEAK's involvement is prominent in angiogenesis inflammatory responses and tissue remodeling suggesting its significance in these biological processes.

Pathways

TWEAK has significant influence on the Nuclear Factor kappa-light-chain-enhancer of activated B cells (NF-kB) and mitogen-activated protein kinases (MAPK) pathways. The interaction of TWEAK with NF-kB can enhance the expression of pro-inflammatory cytokines while its effect on MAPK regulates cell survival and death. TWEAK closely relates to proteins such as TNF-alpha in these pathways often working together to modulate immune responses and cellular environments.

TWEAK is associated with conditions such as rheumatoid arthritis and multiple sclerosis. Its elevated levels have been observed in the synovial tissues of patients with rheumatoid arthritis suggesting its potential role in promoting inflammation. Similarly in multiple sclerosis TWEAK contributes to neuroinflammatory activities. TNF-alpha also links to TWEAK through these diseases showing that both proteins often interact in inflammatory situations which may suggest possible targets for therapeutic interventions.
