Product details

Mouse TYRO3 ELISA Kit is designed for the quantitative determination of TYRO3 in cell culture supernatants, plasma and serum samples.

∆ Note: Mouse TYRO3 concentration is low in normal serum and plasma samples, and may not be detectable in this assay.

This assay employs an antibody specific for mouse TYRO3 coated on a 96-well plate. Standards and samples are pipetted into the wells and TYRO3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse TYRO3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TYRO3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

This ELISA kit shows no cross-reactivity with the following cytokines tested: Mouse CD30, CD40, CRG-2, CTACK, CXCL16, Eotaxin , Eotaxin-2, Fas Ligand, Fractalkine, GCSF, GM-CFS, IFN-gamma, IGFBP-3, IGFBP-5, IGFBP-6, IL-1 alpha, IL-1 beta, IL-2, IL-3, IL-3 Rb, IL-4, IL-5, IL-9, IL-10, IL-12 p40/p70, IL-12 p70, IL-13, IL-17, KC, Leptin R, LEPTIN(OB), LIX, L-Selectin, Lymphotactin, MCP-1, MCP-5, M-CSF, MIG, MIP-1 alpha, MIP-1 gamma, MIP-2, MIP-3 alpha, MIP-3 beta, PF-4, P-Selectin, RANTES, SCF, SDF-1 alpha, TARC, TCA-3, TECK, TIMP-1, TNF-alpha, TNF RI, TNF RII, TPO, VCAM-1, VEGF.

Supplementary information

TYRO3 also known as Tyrosine-protein kinase receptor TYRO3 is a membrane protein involved in various cellular processes. The protein has a molecular mass of approximately 104 kDa. TYRO3 expresses in a range of tissues with high levels noted in the central nervous system reproductive system and immune cells. The protein is part of the TAM family of receptor tyrosine kinases which includes AXL and MERTK and it participates in binding to specific ligands for signal transmission.
Biological function summary

TYRO3 influences multiple cellular activities such as cell survival proliferation and immune regulation. It acts within a signaling complex that includes ligands like Gas6 and Protein S. TYRO3 plays roles in phagocytosis of apoptotic cells and regulation of inflammatory responses. The protein's signaling pathways support tissue homeostasis and modulate immune responses making it an important player in maintaining cellular balance.

Pathways

TYRO3 is engaged in the PI3K/AKT and MAPK pathways which facilitate cell cycle progression and survival. In these pathways it interacts with other proteins such as AXL MERTK and downstream effectors like AKT and ERK. These interactions ensure proper cellular functions with TYRO3-mediated pathways contributing to normal cellular homeostasis and protective responses against stress.

Disruptions in TYRO3 function associate with cancer and autoimmune diseases. Its overexpression or mutations can lead to abnormal cell growth linking TYRO3 to certain cancers. The protein also intersects with immune-related disorders where an imbalance in TYRO3 signaling contributes to diseases like systemic lupus erythematosus. Proteins like AXL and MERTK closely related to TYRO3 often share these pathological associations emphasizing the interconnected nature of the TAM receptor family in disease pathogenesis.

Product protocols

Target data

Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to several ligands including TULP1 or GAS6. Regulates many physiological processes including cell survival, migration and differentiation. Ligand binding at the cell surface induces dimerization and autophosphorylation of TYRO3 on its intracellular domain that provides docking sites for downstream signaling molecules. Following activation by ligand, interacts with PIK3R1 and thereby enhances PI3-kinase activity. Activates the AKT survival pathway, including nuclear translocation of NF-kappa-B and up-regulation of transcription of NF-kappa-B-regulated genes. TYRO3 signaling plays a role in various processes such as neuron protection from excitotoxic injury, platelet aggregation and cytoskeleton reorganization. Also plays an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response by activating STAT1, which selectively induces production of suppressors of cytokine signaling SOCS1 and SOCS3.
