Product details

Abcam's PGF High Sensitivity in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of PGF in milk, culture supernatants, saliva, urine, serum and plasma.

A donkey anti-sheep IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-PGF antigen and a polyclonal sheep antibody specific to PGF. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of PGF captured in the plate.

Prostaglandin F (PGF) is formed in a variety of cells from PGH2, which itself is synthesized from arachidonic acid by the enzyme prostaglandin synthetase. PGF is often viewed as an antagonist to PGE2 due to their opposing effects on various tissues. PGF is a potent bronchoconstrictor and has been implicated in asthma attacks. PGF is also involved in reproductive functions including corpus luteum regulation, uterine contractions, and sperm motility. This has led to its use in terminating pregnancies and inducing labor at term. High levels of PGF have also been associated with preeclampsia.

PGF2 alpha also known as prostaglandin F2α is a prostaglandin with a molecular weight of about 354.49 g/mol. It plays a role in various physiological processes. This compound expresses primarily in the uterus where it acts to induce uterine contractions. It also appears in the corpus luteum and other tissues such as the lungs and kidneys where it has different functions. Researchers frequently use PGF2 alpha ELISA as a method to measure its levels in biological fluids due to the precision and sensitivity of this assay.
Biological function summary

PGF2 alpha participates in reproductive and inflammatory responses. Its primary function involves luteolysis the process of corpus luteum regression necessary for the estrous cycle's progression. PGF2 alpha also promotes smooth muscle contraction including influencing bronchoconstriction and vasoconstriction. Furthermore it functions independently and does not require a complex to execute these actions although other prostaglandins may modulate its effects in specific contexts.

Pathways

PGF2 alpha sits within the prostaglandin signaling pathway which forms a part of the larger arachidonic acid cascade. It interacts with receptors like the FP receptor to mediate its physiological effects. PGF2 alpha also plays a role in the inflammatory pathway where it acts alongside prostaglandin E2 (PGE2) as part of the body's response to injury and infection. Through these signaling networks PGF2 alpha influences other proteins such as cyclooxygenase (COX) enzymes responsible for prostaglandin synthesis.

PGF2 alpha relates significantly to reproductive health issues and inflammatory conditions. For example its excessive activity can lead to dysmenorrhea characterized by painful menstrual cramps as it induces excessive uterine contractions. PGF2 alpha is also linked to asthma where its role in bronchoconstriction contributes to airway narrowing. In both cases the prostaglandin interacts with proteins like COX-2 which upregulates its production during inflammatory and reproductive processes.
