Phospho-c-Jun (S63) and Total c-Jun ELISA Kit (ab279844) is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in mouse cell lysates. By determining phosphorylated c-Jun protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of, mouse phospho-c-Jun and total c-Jun. An antipan c-Jun antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and c-Jun present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho-c-Jun (S63) antibody is added to detect phosphorylated c-Jun. In the remaining wells, rabbit anti-pan-c-Jun antibody is used to detect pan c-Jun. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of c-Jun (S63) or pan c-Jun bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
C-Jun impacts cellular activities by being a significant part of the AP-1 transcription factor complex interacting with other proteins such as Fos. This interaction allows c-Jun to modulate gene expression in response to cellular stimuli. In particular c-Jun influences cell cycle progression and differentiation. Therefore its activity is necessary for physiological and pathological processes. As c-jun's function remains tightly regulated any changes can have wide-ranging effects.
Pathways
C-Jun significantly influences the MAPK/ERK and JNK pathways. These pathways play important roles in cellular responses to stress growth factors and cytokines. Within these pathways c-Jun interacts with proteins such as JNK which phosphorylates c-Jun and enhances its activity. These interactions allow c-Jun to regulate target genes involved in important cellular processes without much delay. Understanding c-Jun's function in these pathways helps elucidate how cells control division survival and apoptosis.
