Product details

Phospho-c-Jun (S63) ELISA Kit (ab279843) is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in mouse cell lysates. By determining phosphorylated c-Jun protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of mouse phospho-c-Jun. An anti-pan c-Jun antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and c-Jun present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-c-Jun (S63) antibody is used to detect phosphorylated c-Jun. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of c-Jun (S63) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The protein c-Jun is an important component of the AP-1 transcription factor complex and is often referred to by alternative names such as c-jun. It has a molecular weight of about 39 kDa. Scientists have found c-jun expressed in a wide range of tissues indicating its diverse roles in cellular functions. This protein plays a critical mechanical role by binding specific DNA sequences to regulate the transcription of various genes involved in cell proliferation and apoptosis.
Biological function summary

C-Jun impacts cellular activities by being a significant part of the AP-1 transcription factor complex interacting with other proteins such as Fos. This interaction allows c-Jun to modulate gene expression in response to cellular stimuli. In particular c-Jun influences cell cycle progression and differentiation. Additionally c-Jun involvement in disorders like Alzheimer's disease stems from its role in apoptotic pathways which may cause neuronal death. Heterodimerizes with proteins of the FOS family to form an AP-1 transcription factor complex, thereby enhancing its DNA binding activity to the AP-1 consensus sequence 5'-TGA[GC]TCA-3' and enhancing its transcriptional activity (PubMed : 2498083).
