Phospho-eIF4EBP1 (T36) and Total eIF4EBP1 ELISA Kit (ab279726) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated eIF4EBP1 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-eIF4EBP1 and total eIF4EBP1. An anti pan eIF4EBP1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and eIF4EBP1 present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho eIF4EBP1 (Thr36) antibody is added to detect phosphorylated eIF4EBP1. In the remaining wells, biotinylated anti-pan-eIF4EBP1 antibody is used to detect pan eIF4EBP1. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG or HRP-Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of eIF4EBP1 (Thr36) or pan eIF4EBP1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
EIF4EBP1 plays an important role in regulating cell growth and proliferation by modulating protein synthesis. It is a part of the eIF4F complex which is responsible for the initiation of mRNA translation. When hypophosphorylated eIF4EBP1 binds tightly to eIF4E and prevents the assembly of the active eIF4F complex leading to reduced translation initiation of mRNAs involved in growth and survival. This regulation is important in conditions where cells need to adapt to metabolic stress or external signals.
Pathways
The mammalian target of rapamycin (mTOR) pathway regulates eIF4EBP1 through phosphorylation. In response to growth signals mTOR phosphorylates eIF4EBP1 causing the release of eIF4E and allowing mRNA translation to proceed. This interaction links eIF4EBP1 to the PI3K/AKT/mTOR signaling pathway which influences cell cycle progression and survival. Related proteins in this pathway include ribosomal protein S6 kinase (S6K1) which is also phosphorylated by mTOR to promote protein synthesis.
