Phospho-GSK3 alpha (S21) and Total GSK3 alpha ELISA Kit (ab279813) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in mouse cell lysates. By determining phosphorylated GSK3 alpha protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of mouse phospho-GSK3 alpha and total GSK3 alpha. An anti-pan GSK3 alpha antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and GSK3 alpha present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho GSK3 alpha (S21) antibody is added to detect phosphorylated GSK3 alpha. In the remaining wells, rabbit anti-pan-GSK3 alpha antibody is used to detect pan GSK3 alpha. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of GSK3 alpha (S21) or pan GSK3 alpha bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

GSK3 alpha influences several cellular functions including cell division differentiation and apoptosis. It functions independently or as part of a protein complex often forming dimeric complexes with GSK3 beta. GSK3 alpha alters the activity of numerous transcription factors and regulates insulin and Wnt signaling pathways affecting glucose metabolism and cellular growth.
Pathways
GSK3 alpha is involved in the Wnt and insulin signaling pathways. In the Wnt pathway GSK3 alpha phosphorylates beta-catenin marking it for degradation and controlling gene expression. In the insulin pathway it modulates glycogen synthesis and affects glucose homeostasis. GSK3 alpha interacts closely with proteins like APC and glycogen synthase making it integral to these pathways.
