Phospho-Phospholipase C gamma 1/PLC-gamma-1 (Y771) ELISA Kit (ab279892) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated C gamma 1/PLC-gamma-1 (Y771) protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-C gamma 1/PLC-gamma-1. An anti-pan C gamma 1/PLC-gamma-1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and C gamma 1/PLC-gamma-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-C gamma 1/PLC-gamma-1 (Y771) antibody is used to detect phosphorylated C gamma 1/PLC-gamma-1. After washing away unbound antibody, HRP conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of C gamma 1/PLC-gamma-1 (Y771) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
PLC-gamma-1 is integral in signal transduction and is an important player in the immune response. The protein does not function as part of a large multi-protein complex but interacts transiently with other signaling molecules. It acts downstream of growth factor receptors and immunoreceptors by mediating calcium release and activation of protein kinase C influencing cell proliferation migration and differentiation. Its expression pattern suggests a critical role in the nervous and immune systems affecting learning memory and immune defense.
Pathways
PLC-gamma-1 is an important component in both the phospholipase C and PLC-gamma pathways. These pathways include important interactions with proteins such as the receptor tyrosine kinases and Src family kinases. In these pathways PLC-gamma-1 modulates signals that guide cellular responses to external stimuli affecting processes like growth signaling and T-cell receptor activation which is vital in adaptive immunity.
