Product details

Phospho-PKMYT1 (T495) and Total PKMYT1 ELISA Kit (ab279891) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated PKMYT1 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-PKMYT1 and total PKMYT1. An anti-pan PKMYT1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and PKMYT1 present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho PKMYT1 (T495) antibody is added to detect phosphorylated PKMYT1. In the remaining wells, biotinylated anti-pan-PKMYT1 antibody is used to detect pan PKMYT1. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG or HRP-Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of PKMYT1 (T495) or pan PKMYT1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

PKMYT1 also known as Membrane-associated tyrosine- and threonine-specific cdc2-inhibitory kinase is an important regulator of the cell cycle. It is a protein kinase with a molecular mass of about 62 kDa and is expressed in various tissues including the brain and reproductive organs. PKMYT1 has a role in regulating the entry into the mitotic phase by inhibiting the Cyclin-dependent kinase 1 (CDK1) which controls the progression through different phases of the cell cycle.
Biological function summary

This kinase is involved in controlling cell division essential for maintaining genomic integrity. PKMYT1 although not always a direct part of a physical complex significantly impacts the activation of CDK1-cyclin B complex which initiates mitosis. By phosphorylating the CDK1 complex PKMYT1 ensures proper timing of cell cycle transitions preventing premature or erroneous mitotic entry.

Pathways

PKMYT1 involvement is notable in the cell cycle regulation specifically during the G2/M transition. PKMYT1 phosphorylates CDK1 inhibiting its activity until the cell is ready to progress into mitosis. Related proteins in this pathway include CDC25 a phosphatase that removes the inhibitory phosphate groups from CDK1 allowing progression to mitosis. Through these interactions PKMYT1 tightly regulates the commitment to mitosis ensuring cells divide accurately.

Studies have linked PKMYT1 to various types of cancer due to its role in cell cycle control. Aberrations in PKMYT1 activity can lead to uncontrolled cell proliferation seen in cancers such as glioblastoma and ovarian cancer. PKMYT1 through its interactions with CDK1 and other cell cycle regulators has potential therapeutic implications. Inhibitors targeting PKMYT1 might help control tumor growth by restoring proper cell cycle regulation serving as a strategic component in cancer therapy.
