Phospho-PYK2 (Y402) and Total PYK2 ELISA Kit (ab279902) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated PYK2 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-PYK2 and total PYK2. An anti-pan PYK2 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and PYK2 present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, mouse anti-phospho PYK2 (Y402) antibody is added to detect phosphorylated PYK2. In the remaining wells, biotinylated anti-pan-PYK2 antibody is used to detect pan PYK2. After washing away unbound antibody, HRP-conjugated anti-mouse IgG or HRP-Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of PYK2 (Y402) or pan PYK2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
PYK2 interacts with integrins and G-protein-coupled receptors (GPCRs) linking extracellular signals to intracellular pathways. It forms complexes with other signaling molecules including Rac1 and c-Src facilitating downstream signaling. These interactions contribute to cellular communication and adhesion processes especially in the central nervous system and immune cells. Through its involvement in signal transduction PYK2 supports cellular responses to environmental changes.
Pathways
PYK2 participates in the MAPK and JNK signaling pathways. These pathways are critical for transmitting signals from the cell surface to the nucleus influencing cellular growth and stress responses. PYK2 interacts closely with other proteins such as Src and FAK which help coordinate signals that regulate cellular dynamics. Through these pathways PYK2 modulates processes involved in the structural reorganization of cells and tissues.
