Phospho-RPS6 (S235/S236) ELISA Kit (ab279912) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in mouse cell lysates. By determining phosphorylated RPS6 (S235/S236) protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of mouse phospho-RPS6. An anti-pan RPS6 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and RPS6 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-RPS6 (S235/S236) antibody is used to detect phosphorylated RPS6. After washing away unbound antibody, HRP conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RPS6 (S235/S236) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

RPS6 also known as Ribosomal Protein S6 is a component of the 40S ribosomal subunit with a molecular weight of approximately 28 kDa. This protein mainly localizes in the cytoplasm where ribosomes are fixed during protein synthesis. The ribosomal machinery uses RPS6 to translate mRNA into polypeptides operating as an essential part of protein biosynthesis. It expresses in various tissues with higher levels in highly proliferative cells which need extensive protein production for growth.
Biological function summary

RPS6 is integral to the formation of the ribosomal complex. This complex is essential for the translation initiation process. RPS6 controls the translation capacity of ribosomes and functions in growth and development by regulating protein synthesis rates. Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome (By similarity).
