Phospho-SHP-1 (S591) ELISA Kit (ab279923) is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated SHP-1 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-SHP-1. An anti-pan SHP-1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and SHP-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and rabbit anti-phospho-SHP-1 (S591) antibody is used to detect phosphorylated SHP-1. After washing away unbound antibody, HRP conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SHP-1 (S591) bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

SHIP-1 can operate independently or as part of larger signaling complexes with other molecules to ensure immune cells respond appropriately to stimuli.
Pathways
SHIP-1 plays an integral role in the phosphoinositide signaling pathway and is linked to the PI3K-Akt pathway. It acts as an important regulatory node by limiting the accumulation of PIP3 a necessary component for Akt activation. This regulation allows for controlled signal propagation that influences immune cell activity and development. SHIP-1 also interfaces with proteins like PTEN and SHP-1 which similarly modulate downstream signaling events within these pathways ensuring the maintenance of immune equilibrium.
