Product details

Phospho-TAK1 (S412) and Total TAK1 ELISA Kit (ab279951) is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated TAK1 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-TAK1 and total TAK1. An anti-pan TAK1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and TAK1 present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho TAK1 (S412) antibody is added to detect phosphorylated TAK1. In the remaining wells, biotinylated anti-pan-TAK1 antibody is used to detect pan TAK1. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG or HRP-Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TAK1 (S412) or pan TAK1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

TAK1 also known as MAP3K7 (Mitogen-activated protein kinase kinase kinase 7) is an important protein kinase that weighs approximately 63 kDa. This protein is expressed in various tissues and cells including HEK 293T cells. TAK1 plays a mechanical role as a part of the MAPK signaling cascade. It phosphorylates and activates downstream kinases which is key to transmitting cellular signals that regulate responses to external stimuli like cytokines and stress.
Biological function summary

TAK1 is involved in several cellular processes necessary for maintaining balance and responding to stress. TAK1 forms a complex with proteins like TAB1 TAB2 and TAB3 which are important for its activation and signaling function.
