Phospho-Eph receptor B2 ELISA Kit (ab279783) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated Eph receptor B2 protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-Eph receptor B2. An anti-Eph receptor B2 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and phosphorylated and unphosphorylated Eph receptor B2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-phosphotyrosine antibody is used to detect only tyrosine-phosphorylated protein. After washing away unbound antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of phospho-Eph receptor B2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Eph receptor B2 also known as EphB2 is a member of the Eph receptor family which includes a number of receptor tyrosine kinases. It has a molecular weight of approximately 110 kDa. The B2 receptor is strongly expressed in various tissues including the brain spinal cord and developing embryos. These proteins include the Eph protein as well as other B2 proteins. The receptor plays a mechanical role in guiding cell positioning and tissue patterning by interacting with Ephrin ligands specifically ephrin-B2 and ephrin-B3.
Biological function summary

Eph receptor B2 functions in cell-cell communication and is involved in neuronal navigation and synaptic plasticity. It forms part of a complex that brings together ligand-receptor pairs on adjacent cells facilitating bidirectional signaling. The receptor's activation influences neuronal circuitry remodeling and helps in the differentiation of numerous cell types. In Alzheimer's disease changes in B2 receptor signaling are associated with disrupted synaptic plasticity leading to cognitive deficits highlighting the importance of the Eph B2 receptor and its interplay with related signaling proteins in disease pathways.
