Phosphotyrosine GM-CSF Receptor alpha ELISA Kit (ab279858) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated GM-CSF Receptor alpha protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-GM-CSF Receptor alpha. An anti-pan GM-CSF Receptor alpha antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and phosphorylated and unphosphorylated GM-CSF Receptor alpha present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-phosphotyrosine antibody is used to detect only tyrosine-phosphorylated protein. After washing away unbound antibody, HRP-conjugated Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of phospho-GM-CSF Receptor alpha bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
GM-CSF receptor alpha binds GM-CSF with high affinity and is part of a receptor complex that includes beta common chain (βc). The receptor complex initiates signaling cascades that modulate immune functions such as differentiation of myeloid cells and macrophage activation. These interactions support various immune processes like inflammation and pathogen defense. GM-CSF receptor alpha is essential to ensure proper cellular responses to environmental cues.
Pathways
GM-CSF receptor alpha is involved in the JAK-STAT and MAPK pathways. Upon ligand binding it activates JAK2 which in turn triggers STAT5 phosphorylation — important for transcription of immune-related genes. Simultaneously it initiates the MAPK (mitogen-activated protein kinase) pathway further promoting cell differentiation and proliferation. Each of these signaling events connect GM-CSF receptor alpha to other proteins like JAK2 and STAT5 which collaborate to sustain proper immune activity.
