Phosphotyrosine ITK/EMT ELISA Kit (ab279835) is a very rapid, convenient and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated ITK/EMT protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.

This Sandwich ELISA kit is an in vitro enzyme-linked immunosorbent assay for the measurement of human phospho-ITK/EMT. An anti-pan ITK/EMT antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and phosphorylated and unphosphorylated ITK/EMT present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-phosphotyrosine antibody is used to detect only tyrosine-phosphorylated protein. After washing away unbound antibody, HRP-conjugated Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of phospho-ITK/EMT bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Activity summary

ITK also known as Interleukin-2-inducible T-cell kinase is a protein belonging to the Tec family of non-receptor tyrosine kinases. It weighs approximately 72 kDa and plays a significant role in signal transduction in immune cells specifically T cells. ITK is widely expressed in lymphoid tissues particularly in thymocytes and peripheral T cells. ITK products often include variants like ITK-1 and ITK kit which are useful in various scientific applications.

Biological function summary

ITK is integral to the activation and development of T cells which are critical components of the adaptive immune response. ITK functions as part of a larger signaling complex interacting with proteins such as LAT and SLP-76 to mediate downstream signaling events following T-cell receptor engagement.
