Recombinant Prostaglandin E2 was spiked into the following samples and diluted in a 2-fold dilution series in Sample Diluent PTR. Undiluted samples are 0.5 mg/mL Hela cell extract and Jurkat cell extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Prostaglandin E2 SimpleStep ELISA® kit is a single-wash 90 min competitive ELISA designed for the quantitative measurement of Prostaglandin E2 in Cell Lysate and Tissue Extract. Quantitate Prostaglandin E2 with 6.88 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture conjugates conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to competitive ELISA allows the formation of the antibody-analyte complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies

Prostaglandin E2 (PGE2) also known as dinoprostone is a lipid compound with a molecular weight of approximately 352.47 g/mol. This compound is synthesized in various tissues like the kidneys lungs and brain primarily by the enzymatic action of cyclooxygenase (COX) on arachidonic acid. As a member of the prostaglandin family PGE2 functions as an important signaling molecule and mediator of inflammation. Researchers often use PGE2 ELISA kits and assays including those specific for mouse PGE2 to measure its levels accurately in biological samples.
Biological function summary

As a potent mediator PGE2 plays roles in inflammation fever induction and regulation of vascular tone. It exerts its effects through specific G-protein-coupled receptors (EP1 EP2 EP3 and EP4) on the surface of target cells. Although not part of a strict protein complex PGE2 works in conjunction with other inflammatory mediators including cytokines and other prostaglandins. It influences cellular activities by modulating the intracellular cyclic AMP levels and calcium fluxes.

Pathways

PGE2 is an important component of the prostaglandin biosynthesis pathway and the broader inflammatory signal transduction pathway. Within these pathways it relates to the COX enzymes specifically COX-2 which converts arachidonic acid into PGH2 the precursor of PGE2. This metabolic pathway highlights the interplay between PGE2 and other prostanoids impacting inflammatory responses and homeostatic processes.

PGE2 has associations with conditions such as arthritis and cancer. Elevated levels of PGE2 are often found in inflamed tissues and have been linked to the pathogenesis of rheumatoid arthritis by promoting inflammation and pain. In cancer PGE2 can encourage tumoral growth by modulating the immune response and promoting angiogenesis. In these contexts the interactions of PGE2 with enzymes like COX-2 and cytokines such as tumor necrosis factor (TNF) further connect its roles to disease progression and potential therapeutic targets.
