Protein Disulfide Isomerase/P4HB ELISA Kit - Intracellular is a single-wash 90-min Simplestep used to quantify Protein Disulfide Isomerase/P4HB with a sensitivity of 45.9pg/ml. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

Interpolated concentration of native Protein Disulfide Isomerase/P4HB was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1.25 µg/mL MCF7 cell extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer SSW.

How the assay works

Protein Disulfide Isomerase/P4HB ELISA Kit - Intracellular (ab324925) SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. This multifunctional protein catalyzes the formation, breakage and rearrangement of disulfide bonds. At the cell surface, seems to act as a reductase that cleaves disulfide bonds of proteins attached to the cell. May therefore cause structural modifications of exofacial proteins. Inside the cell, seems to form/rearrange disulfide bonds of nascent proteins. At high concentrations and following phosphorylation by FAM20C, functions as a chaperone that inhibits aggregation of misfolded proteins (PubMed : 32149426). At low concentrations, facilitates aggregation (anti-chaperone activity). May be involved with other chaperones in the structural modification of the TG precursor in hormone biogenesis. Also acts as a structural subunit of various enzymes such as prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein MTTP. Receptor for LGALS9; the interaction retains P4HB at the cell surface of Th2 T helper cells, increasing disulfide reductase activity at the plasma membrane, altering the plasma membrane redox state and enhancing cell migration (PubMed : 21670307).
