Protein L Ligand Leakage Detection ELISA Kit (ab325035) is a sandwich ELISA for the quantification of ligand leakage in eluates from Protein L affinity chromatography medium containing immunoglobulin (Ig) or Ig-fragments.
How the assay works
Microwells are pre-coated with anti-protein L Ligand antibodies to capture protein L ligand from samples and standards pipetted into the wells. Following binding, an anti protein L ligand-HRP conjugate is added and incubated. After washing to remove non-specific interactions, a ready-to-use TMB substrate is introduced, triggering a colorimetric reaction proportional to the concentration of protein L ligand in the sample. The reaction is halted by adding a stop solution, and absorbance is measured at 450 nm.

1. Add 50µL of standard or diluted samples to appropriate wells. Incubate for 60 min at room temperature
2. Wash
3. Add 100µL of Anti-Protein L Ligand :HRP Conjugate to each well. Incubate 60 min at room temperature
4. Wash
5. Add 100µL TMB substrate to each well. Incubate for 30 min in the dark
6. Add 100µL of Stop solution to each well
7. Read at 450 nm with 10-15 min.
