Interpolated concentrations of native Albumin in rat serum and plasma samples.
The concentrations of Albumin were measured in duplicates, interpolated from the Albumin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 2 million, plasma (heparin) 1 : 8 million, plasma (EDTA) 1 : 4 million, and plasma (citrate) 1 : 4 million. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Albumin concentration in the neat samples was determined to be 32 mg/mL in serum, 29 mg/mL in plasma (heparin), 26 mg/mL in plasma (EDTA), and 23 mg/mL in plasma (citrate).

The concentrations of Albumin were measured in duplicate and interpolated from the Albumin standard curve and corrected for sample dilution. Undiluted samples are 1 : 1,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Albumin concentration in the neat sample was determined to be 18 μg/mL.

How the assay works
Rat Albumin SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Biological function summary
Albumin plays a critical role in transporting small molecules like metabolites and drugs through the circulatory system. It is not a part of any complex but functions independently to bind transport and release various ligands. Albumin's affinity for hydrophobic molecules makes it essential for they to be solubilized in blood for proper metabolism and excretion.
Pathways
More than five binding sites on albumin facilitate its role in the fatty acid metabolism and renin-angiotensin system pathways. In fatty acid metabolism albumin interacts with transport proteins carrying fatty acids to tissues for energy production. Additionally during the renin-angiotensin system albumin modulates blood pressure through its interaction with angiotensin II which can influence vasoconstriction.
