Interpolated concentrations of native Beta-2-Microglobulin in rat urine and cell culture supernatant samples.
The concentrations of Beta-2-Microglobulin were measured in duplicates, interpolated from the Beta-2-Microglobulin standard curves and corrected for sample dilution. Undiluted samples are as follows : urine 1 : 150 and lung cell culture supernatant 1 : 100. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Beta-2-Microglobulin concentration was determined to be 491.5 ng/mL in neat rat urine and 434.3 ng/mL in neat rat lung cell culture supernatant.

Interpolated concentrations of native Beta-2-Microglobulin in rat serum and plasma samples.
The concentrations of Beta-2-Microglobulin were measured in duplicates, interpolated from the Beta-2-Microglobulin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 3,000, plasma (citrate) 1 : 1,500, plasma (heparin) 1 : 2,250, and plasma (EDTA) 1 : 2,250. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Beta-2-Microglobulin concentration was determined to be 12.23 μg/mL in neat serum, 7.025 μg/mL in neat plasma (citrate), 8.710 μg/mL in neat plasma (heparin), and 7.155 μg/mL in neat plasma (EDTA).

Rat Beta-2-Microglobulin ELISA Kit (ab264619) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Beta-2-Microglobulin protein in cell culture media, cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat Beta-2-Microglobulin with 268 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Beta-2-Microglobulin is the light chain of the class I major histocompatibility complex (MHC); it forms heterodimers with classical (HLA-A, -B, -C) and non-classical (HLA-E, -F, -G) class I MHC heavy chains and with class I non-MHC heavy chains (CD1). Beta-2-Microglobulin is involved in the presentation of peptide antigens to the immune system. Beta-2-Microglobulin can dissociate from the MHC and circulate in complex with alpha 2-Macroglobulin.

Biological function summary
Beta-2-microglobulin is important for the stability and transport of MHC class I molecules to the cell surface. As part of the MHC class I complex B2M assists in binding peptides allowing immune cells to identify and target pathogen-infected cells. Without B2M the MHC class I molecules are not properly expressed on the cell surface disrupting immune recognition. In laboratory settings researchers often use anti-beta-2-microglobulin antibodies to investigate its role in MHC class I function.
Pathways
Beta-2-microglobulin interacts significantly with the immune system most notably in the antigen processing and presentation pathway. It works alongside proteins such as the heavy chain of MHC class I. B2M is important in the pathway that involves the transport of antigens to the endoplasmic reticulum where they are loaded onto MHC class I molecules for inspection by cytotoxic T cells. Another related pathway is the tapasin-mediated processing of antigen peptides highlighting the indispensable role of B2M in immune response regulation.
