Abcam's beta NGF Rat ELISA Kit (ab193736) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat beta NGF in cell lysate and tissue lysate.
This assay employs an antibody specific for rat beta NGF coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures beta NGF present in the samples. The wells are washed and biotinylated anti-rat beta NGF antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of beta NGF bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
Beta nerve growth factor plays an important role in the growth survival and differentiation of neurons. As a member of the neurotrophin family it binds to receptors like TrkA and p75NTR on the surface of nerve cells. This binding activity helps to promote neuronal health and function especially in the peripheral and central nervous systems. While beta-NGF traditionally acts independently it also interacts with other signaling molecules to trigger cascades that support cell survival and neural connections.
Pathways
Beta-NGF prominently participates in the MAPK/ERK and PI3K/AKT signaling pathways. These pathways are essential for transmitting signals from the cell surface to the DNA in the nucleus therefore regulating gene expression linked to cell growth and survival. In these contexts beta-NGF works closely with proteins like Akt and ERK enhancing neuron resilience and promoting restorative mechanisms across the nervous system.
