Product details

Abcam's betaNGF Rat ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat betaNGF in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Rat betaNGF coated on a 96-well plate. Standards and samples are pipetted into the wells and betaNGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat betaNGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of betaNGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Beta nerve growth factor (beta-NGF) is a significant protein that functions in the development and maintenance of the nervous system. It is also known as NGF or NGFB and has a molecular mass of around 26 kDa. This protein is highly expressed in a wide range of tissues including the brain skin and various glands. Researchers often use assays like growth factor assays and NGF ELISA kits to measure beta-NGF levels given its importance in several physiological processes.
Biological function summary

Beta nerve growth factor plays an important role in the growth survival and differentiation of neurons. As a member of the neurotrophin family it binds to receptors like TrkA and p75NTR on the surface of nerve cells. This binding activity helps to promote neuronal health and function especially in the peripheral and central nervous systems. While beta-NGF traditionally acts independently it also interacts with other signaling molecules to trigger cascades that support cell survival and neural connections.

Nerve growth factor is important for the development and maintenance of the sympathetic and sensory nervous systems. Extracellular ligand for the NTRK1 and NGFR receptors, activates cellular signaling cascades to regulate neuronal proliferation, differentiation and survival (By similarity). The immature NGF precursor (proNGF) functions as a ligand for the heterodimeric receptor formed by SORCS2 and NGFR, and activates cellular signaling cascades that lead to inactivation of RAC1 and/or RAC2, reorganization of the actin cytoskeleton and neuronal growth cone collapse. In contrast to mature NGF, the precursor form (proNGF) promotes neuronal apoptosis (in vitro) (By similarity). Inhibits metalloproteinase-dependent proteolysis of platelet glycoprotein VI (By similarity). Binds lysophosphatidylinositol and lysophosphatidylserine between the two chains of the homodimer. The lipid-bound form promotes histamine relase from mast cells, contrary to the lipid-free form (By similarity).
