Abcam's Fas Ligand (CD95L) Rat ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat Fas Ligand (CD95L) in plasma and cell culture supernatants. (Rat Fas Ligand concentration is pretty low in normal plasma, it may not be detected in this assay).

This assay employs an antibody specific for Rat Fas Ligand coated on a 96-well plate. Standards and samples are pipetted into the wells and Fas Ligand present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat Fas Ligand antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Fas Ligand bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

We have not been able to detect endogenous Rat Fas Ligand in normal serum with ab100759, only in serum spiked with Rat Fas Ligand.

Target data

Cytokine that binds to TNFRSF6/FAS, a receptor that transduces the apoptotic signal into cells (PubMed : 7505205). Involved in cytotoxic T-cell-mediated apoptosis, natural killer cell-mediated apoptosis and in T-cell development (PubMed : 7505205). Initiates fratricidal/suicidal activation-induced cell death (AICD) in antigen-activated T-cells contributing to the termination of immune responses (By similarity). TNFRSF6/FAS-mediated apoptosis also has a role in the induction of peripheral tolerance (By similarity). Binds to TNFRSF6B/DcR3, a decoy receptor that blocks apoptosis (By similarity).. Tumor necrosis factor ligand superfamily member 6, soluble form. Induces FAS-mediated activation of NF-kappa-B, initiating non-apoptotic signaling pathways. Can induce apoptosis but does not appear to be essential for this process.. FasL intracellular domain. Cytoplasmic form induces gene transcription inhibition.
