Sandwich ELISA - Rat FGF-21 ELISA Kit (AB223589)
Interpolated concentrations of native FGF-21 in rat serum and plasma samples.
The concentrations of FGF-21 were measured in duplicates, interpolated from the FGF-21 standard curves and corrected for sample dilution. Undiluted samples are as follows : normal serum neat, pregnant serum 12.5%, normal plasma (citrate) neat, normal plasma (EDTA) neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean FGF-21 concentration was determined to be 580.1 pg/mL in neat normal serum, 3361 pg/mL in neat pregnant serum, 548.7 pg/mL in neat normal plasma (citrate), and 782.4 pg/mL in neat normal plasma (EDTA).

Product details
Rat FGF-21 ELISA Kit (ab223589) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of FGF-21 protein in cell culture supernatant, cit plasma, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat FGF-21 with 6.8 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
FGF-21 is a secreted protein, a member of a large FGF gene family. FGF-21 stimulates glucose uptake in differentiated adipocytes via the induction of glucose transporter SLC2A1/GLUT1 expression but not via SLC2A4/GLUT4 expression. FGF-21 activity probably requires the presence of KLB.

Biological function summary
FGF21 influences glucose and lipid metabolism. It does not act as part of a larger protein complex. Instead it directly affects metabolic processes enhancing insulin sensitivity and lipid oxidation while reducing body weight. These actions help maintain energy balance and homeostasis making FGF21 an important player in metabolic control.
Pathways
FGF21 plays a significant role in the insulin signaling pathway and PPARγ coactivator 1α (PGC-1α) pathway. These pathways regulate cellular energy metabolism and mitochondrial biogenesis respectively. FGF21 interacts with other molecules such as PPARα (peroxisome proliferator-activated receptor alpha) to exert its effects demonstrating its integration into broader metabolic networks. FGF21's interaction with these pathways highlights its function in energy utilization and storage.
