The concentrations of Fractalkine were measured in duplicates, interpolated from the Fractalkine standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (EDTA) 100%, plasma (heparin) 50% and plasma (citrate) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fractalkine concentration was determined to be 151 ng/mL in serum, 118 ng/mL in plasma (EDTA) 250 ng/mL in plasma (heparin), and 93 ng/mL in plasma (citrate).

Rat Fractalkine ELISA Kit (ab229890) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fractalkine protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat Fractalkine with 9.8 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. CX3CL1 also known as fractalkine is a chemokine protein characterized mechanically by its dual function as both a chemoattractant and adhesion molecule. It exhibits a unique membrane-bound structure and can exist in a soluble form after proteolytic cleavage. The mass of CX3CL1 is approximately 42-45 kDa. This protein is highly expressed in endothelial cells neurons and tissues with substantial vascularization. Considerable interest surrounds the potential of CX3CL1 assessment in various studies often involving applications like fractalkine ELISA or assays on mouse models.
Biological function summary

CX3CL1 is involved in the immune system and nervous system functions influencing leukocyte adhesion and migration. It is not known to be part of a large multiprotein complex but directly interacts with its fractalkine receptor CX3CR1 on the surface of immune cells. Chemokine that acts as a ligand for both CX3CR1 and integrins ITGAV : ITGB3 and ITGA4 : ITGB1. The CX3CR1-CX3CL1 signaling exerts distinct functions in different tissue compartments, such as immune response, inflammation, cell adhesion and chemotaxis. Regulates leukocyte adhesion and migration processes at the endothelium. Can activate integrins in both a CX3CR1-dependent and CX3CR1-independent manner. In the presence of CX3CR1, activates integrins by binding to the classical ligand-binding site (site 1) in integrins. In the absence of CX3CR1, binds to a second site (site 2) in integrins which is distinct from site 1 and enhances the binding of other integrin ligands to site 1.. Processed fractalkine. The soluble form is chemotactic for T-cells and monocytes, but not for neutrophils.. Fractalkine. The membrane-bound form promotes adhesion of those leukocytes to endothelial cells.
