Interpolated concentrations of native IL-10 in rat cell culture supernatant samples.

The concentrations of IL-10 were measured in duplicates, interpolated from the IL-10 standard curves and corrected for sample dilution. Undiluted samples are as follows : rat PBMC stimulated with PHA (10 μg/mL, 48 hours) supernatant 50%, unstimulated PBMC supernatant 100%, and spleen stimulated with concanavalin A (5 μg/mL, 48 hours) supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-10 concentration was determined to be 1,174 pg/mL in PBMC stimulated with PHA supernatant, 230 pg/mL in unstimulated PBMC supernatant and 789 pg/mL in spleen stimulated with concanavalin A supernatant.

Rat peripheral blood mononuclear cells were cultured unstimulated or stimulated with 10 µg/mL PHA.

Conditioned media was harvested after 48 hours. IL-10 was measured in 50% unstimulated and PHA stimulated PBMC supernatant. The concentrations of IL-10 were measured in duplicate, interpolated from the IL-1beta standard curves and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-10 concentration was determined to be 1,068 pg/mL in PHA stimulated PBMC supernatant and 227 pg/mL in unstimulated PBMC supernatant.

How the assay works

Rat IL-10 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Target data

Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3. In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha. Interferes also with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation (By similarity). In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling (By similarity).
