Interpolated concentrations of native IL-7 in rat serum.
The concentrations of IL-7 were measured in duplicates, interpolated from the IL-7 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-7 concentration was determined to be 251.84 pg/mL in serum.

Rat IL-7 ELISA kit (ab277704) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Rat IL-7 protein in rat serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-7 with 1.27 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. IL-7 is a cytokine important for B and T cell development. In humans, IL-7 and hepatocyte growth factor form a heterodimer that functions as a pre-pro-B cell growth-stimulating factor. IL-7 is found to be a cofactor for V(D)J rearrangement of the T cell receptor beta (TCRB) during early T cell development. It can be produced locally by intestinal epithelial and epithelial goblet cells and may serve as a regulatory factor for intestinal mucosal lymphocytes. Knockout studies in mice suggested that IL-7 plays an essential role in lymphoid cell survival. Rat IL-7 shares 89% and 59% sequence homology with mouse and human respectively.

Biological function summary
IL7 is essential for the survival and proliferation of lymphoid cells. It acts on both T cells and B cells critical components of the adaptive immune system. The protein supports the survival of naïve and memory T cells playing a significant role in T cell homeostasis. IL7 does not operate as part of a large complex but interacts directly with its receptor to exert its function. Through its influence on T and B cell proliferation the cytokine sustains lymphoid system functionality and integrity.
Pathways
IL7 plays a pivotal role in the JAK/STAT signaling pathway which is central to cellular responses to cytokines. When IL7 binds to its receptor JAK kinases activate and promote the phosphorylation of STAT5 leading to gene transcription necessary for cell survival and proliferation. Another important pathway is the PI3K/AKT signaling cascade where the cytokine supports cell survival through anti-apoptotic signals. Through these pathways IL7 is interconnected with other cytokines such as IL2 that also function through shared or similar signaling cascades contributing to immune homeostasis.
