Sandwich ELISA - Rat IL-9 ELISA Kit (AB252353)
Interpolated concentrations of native IL-9 in rat tissue culture supernatant samples.
Rat spleen tissue was cultured in the presence of 10 ng/mL PMA and 10 ng/mL rat IL-1a for 4 days (PMA + IL-1a) or 50 ng/mL PMA and 500 ng/mL calcium ionomycin for 3 days (PMA + calcium ionomycin). Spleen tissue was cultured without treatment for 2 days (unstimulated). The concentrations of IL-9 were measured in duplicates, interpolated from the IL-9 standard curves and corrected for sample dilution. Undiluted samples are as follows : PMA + IL-1a 100%, PMA + calcium ionomycin 100%, and unstimulated 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-9 concentration was determined to be 91.51 pg/mL in neat PMA + IL-1a stimulated spleen tissue supernatant, 126.3 pg/mL in neat PMA + calcium ionomycin stimulated spleen tissue supernatant, and 19.64 pg/mL in neat unstimulated spleen tissue supernatant.

Rat IL-9 ELISA Kit (ab252353) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL-9 protein in cit plasma, edta plasma, hep plasma, serum, tissue culture media, and cell culture media. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-9 with 7.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
IL-9 influences immune responses and is important for various processes including cell proliferation and survival. IL-9 activates mast cells which play role in allergy response. It also affects the growth and differentiation of T cells and enhances the function of dendritic cells. IL-9 operates individually and not as part of a larger complex but works closely with its receptor IL-9R to initiate downstream signaling events.
