Interpolated concentrations of native L-Selectin in serum, rat plasma (EDTA), and plasma (citrate) samples.

The concentrations of L-Selectin were measured in duplicates, interpolated from the L-Selectin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (EDTA) 100%, and plasma (citrate) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean L-Selectin concentration was determined to be 41.8 ng/mL in serum, 30 ng/mL in plasma (EDTA), and 25.1 ng/mL in plasma (citrate).

Rat L-Selectin ELISA Kit (ab263876) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of L-Selectin protein in cell culture supernatant, cit plasma, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat L-Selectin with 0.77 ng/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. L-Selectin, also referred to as CD62L and Leukocyte adhesion molecule 1, is a calcium-dependent lectin encoded by the gene SELL. L-Selectin binds to glycoproteins to mediate cell adhesion on neighboring cells. In peripheral lymph nodes, L-selectin mediates the adherence of lymphocytes to endothelial cells with high endothelial venules. L-Selectin is composed of several domains, one homologous to lectins, an epidermal growth factor domain, and two to repeat units found In C3/C4 binding proteins.

CD62L also known as L-selectin is a cell adhesion molecule with a mass of approximately 74 kDa. It is part of the L-selectin family and is found on the surface of leukocytes including T cells B cells neutrophils and monocytes. Mouse models frequently study CD62L under the name "MEL-14". CD62L plays an important role in mediating the adhesion and rolling of leukocytes on blood vessel endothelial cells during the immune response. Its structure allows it to interact specifically with carbohydrates on endothelial cells facilitating the initial steps of leukocyte extravasation into tissues.
Biological function summary

CD62L is involved in the migration of leukocytes into lymphoid tissues and sites of inflammation. It is not known to be part of a larger protein complex; rather it functions independently on the cell membrane. CD62L also assists in the homing of naive lymphocytes to peripheral lymph nodes. The molecule regulates how immune cells enter tissues through high endothelial venules acting as a critical entry point for leukocyte trafficking.

Pathways

CD62L contributes significantly to the immune trafficking pathway and leukocyte adhesion cascade. It works closely with other adhesion molecules like CD44 and CD54 (ICAM-1) which are important within the context of cellular responses during inflammation.
