Sandwich ELISA - Rat Leptin ELISA Kit (AB229891)
Interpolated concentrations of spike Leptin in rat cell culture supernatant.
The concentrations of Leptin were measured in duplicates, interpolated from the Leptin standard curves and corrected for sample dilution. Undiluted samples are as follows : cell culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Sandwich ELISA - Rat Leptin ELISA Kit (AB229891)
Interpolated concentrations of native Leptin in rat serum and plasma samples.
The concentrations of Leptin were measured in duplicates, interpolated from the Leptin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Target concentration was determined to be 1889 pg/mL in serum, 1326 pg/mL in plasma (heparin) and 1026 pg/mL in plasma (citrate).

Product details
Rat Leptin ELISA Kit (ab229891) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Leptin protein in cell culture supernatant, cit plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat Leptin with 24 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Biological function summary
Leptin regulates food intake and energy expenditure by binding to its receptors in the hypothalamus. It is an important part of the feedback loop to manage body weight and appetite. Leptin is not part of a larger protein complex and acts directly on its target tissues. By signal transduction pathways it influences cellular processes such as metabolism and even impacts fertility and immune responses.
Pathways
Leptin is involved in the neuroendocrine signaling pathway and the MAPK/ERK pathway. In the neuroendocrine system leptin interacts with proteins like proopiomelanocortin (POMC) and neuropeptide Y (NPY) to manage appetite and energy use. Additionally it activates the JAK/STAT pathway when binding to its receptor which influences gene expression involved in metabolism and inflammation.
