Interpolated concentrations of native Lipocalin-2 in rat serum and plasma samples. The concentrations of Lipocalin-2 were measured in duplicates interpolated from the Lipocalin-2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 500 plasma (EDTA) 1 : 800 plasma (heparin) 1 : 5000 and plasma (citrate) 1 : 13333. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Lipocalin-2 concentration was determined to be 293 ng/mL in serum 290 ng/mL in plasma (EDTA) 560 ng/mL in plasma (heparin) and 4326 ng/mL in plasma (citrate).

Undiluted samples are as follows : urine 1 : 10000 and lung cell culture supernatant 1 : 400. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Lipocalin-2 concentration was determined to be 2184 ng/mL in urine and 297 ng/mL in lung supernatant.

How the assay works

Rat Lipocalin-2 ELISA Kit, Chemiluminescent (ab326403) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Iron-trafficking protein involved in multiple processes such as apoptosis, innate immunity and renal development (By similarity). Binds iron through association with 2,3-dihydroxybenzoic acid (2,3-DHBA), a siderophore that shares structural similarities with bacterial enterobactin, and delivers or removes iron from the cell, depending on the context. Iron-bound form (holo-24p3) is internalized following binding to the SLC22A17 (24p3R) receptor, leading to release of iron and subsequent increase of intracellular iron concentration. In contrast, association of the iron-free form (apo-24p3) with the SLC22A17 (24p3R) receptor is followed by association with an intracellular siderophore, iron chelation and iron transfer to the extracellular medium, thereby reducing intracellular iron concentration. Involved in apoptosis due to interleukin-3 (IL3) deprivation : iron-loaded form increases intracellular iron concentration without promoting apoptosis, while iron-free form decreases intracellular iron levels, inducing expression of the proapoptotic protein BCL2L11/BIM, resulting in apoptosis (By similarity). Involved in innate immunity; limits bacterial proliferation by sequestering iron bound to microbial siderophores, such as enterobactin. Can also bind siderophores from M.tuberculosis (By similarity).
