Interpolated concentrations of native M-CSF in rat serum and plasma samples.
The concentrations of M-CSF were measured in duplicates, interpolated from the M-CSF standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 30%, plasma (citrate) 30%, plasma (heparin) 40% and plasma (EDTA) 40% %. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean M-CSF concentration was determined to be 39,17 pg/mL in serum, 2,688 pg/mL, plasma (citrate), 2,433 pg/mL in plasma (EDTA) and 4,740 pg/mL in plasma (heparin).

Rat M-CSF ELISA Kit (CSF1) (ab253214) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of M-CSF (CSF1) protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat M-CSF (CSF1) with 6 pg/ml sensitivity.

Macrophage colony-stimulating factor 1 (M-CSF), also known as CSF1, is a secreted cytokine that plays an essential role in the regulation of differentiation and maintenance of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. It plays an important role in innate immunity and in inflammatory processes where it acts to promote the release of proinflammatory chemokines. M-CSF also plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. Reports show that it is required for normal male and female fertility. It also acts in reorganization of the actin cytoskeleton, regulates formation of membrane ruffles, cell adhesion and cell migration.

Biological function summary
M-CSF influences the production differentiation and function of macrophages and osteoclasts. It is not part of a large complex but interacts with its receptor the CSF1R (colony-stimulating factor 1 receptor) on the surface of target cells. This interaction promotes the survival and proliferation of the target cells playing significant roles in immune response and bone homeostasis by regulating osteoclast development.
Pathways
The interaction of M-CSF with its receptor is central to several biological pathways notably the immune system pathway and bone resorption pathway. Within these pathways the binding of M-CSF to CSF1R activates downstream signaling cascades such as the PI3K/AKT and MAPK pathways importantly affecting cell survival and differentiation. The M-CSF pathway intersects with other cytokines and factors like GM-CSF (granulocyte-macrophage colony-stimulating factor) which also regulate immune cell dynamics.
