Interpolated concentrations of native MIP2 in rat serum, lung supernatant treated with or without 10 µg/mL Con A for 3 days, and heart supernatant samples.
The concentrations of MIP2 were measured in duplicates, interpolated from the MIP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, lung supernatant treated with Con A 0.5%, lung supernatant mock treated 1.25%, heart supernatant 1.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MIP2 concentration was determined to be 104 pg/mL in neat serum, 35,588 pg/mL in neat Con A treated lung supernatant, 17,197 in neat mock treated lung supernatant and 10,566 pg/mL in neat heart supernatant.

Rat MIP2 ELISA kit (ab282308) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Rat MIP2 protein in rat serum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat MIP2 with 0.37 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Macrophage Inflammatory Protein 2 (MIP2), also known as C-X-C motif chemokine 2 (CXCL2), is a small cytokine belonging to the CXC chemokine family. MIP2 forms homotetramers. MIP2 is produced by activated monocytes and neutrophils and expressed at sites of inflammation. MIP2 is chemotactic for polymorphonuclear leukocytes. Mature chain of rat MIP2 is 79% identical to rat C-X-C Motif Chemokine 3, and 74% identical to rat Growth-Regulated Alpha Protein. Rat MIP2 is 88% identical to mouse MIP2.

Biological function summary
CXCL2 attracts and activates neutrophils as part of the innate immune response. It acts independently not as part of a larger complex to provoke a chemotactic response guiding neutrophils to areas of tissue injury. CXCL2 also promotes the release of other cytokines and enzymes that contribute to inflammation. Its activity is mediated through its interaction with receptors like CXCR2 on target cell surfaces.
Pathways
CXCL2 is involved in inflammatory signaling and leukocyte migration. It holds importance in the chemokine signaling pathway which regulates leukocyte trafficking. CXCL2 interacts with proteins like CXCR2 influencing the inflammation process by activating downstream kinases and other signal transduction molecules such as mitogen-activated protein kinases (MAPKs).
