Sandwich ELISA - Syntaxin 1a ELISA Kit (AB277088)
Interpolated concentrations of native Syntaxin 1a in 80 μg/mL SHSY-5Y cell extract, 20 μg/mL mouse brain tissue extract and 15 μg/mL rat brain tissue extract.
The concentrations of Syntaxin 1a were measured in duplicate and interpolated from the Syntaxin 1a standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Syntaxin 1a concentration was determined to be 3,353.80 pg/mL in SHSY-5Y cell extract, 2,463.89 pg/mL in mouse brain tissue extract and 3,289.96 pg/mL in rat brain tissue extract.

Product details
Syntaxin 1a ELISA kit (ab277088) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human/Mouse/Rat Syntaxin 1a protein in cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Syntaxin 1a with 36 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Syntaxin 1a is part of the SNARE complex, which plays a critical role in neurotransmission. With its partners, SNAP25 and VAMP2, it is responsible for regulating calcium ion channels and for fusion of synaptic vesicles with the presynaptic membrane. This protein shares homology across Mammalia, and therefore is expected to cross-react with many species. The antibodies in this kit were raised against the human full-length recombinant protein.

Biological function summary
The involvement of Syntaxin 1a in synaptic transmission is significant. It forms a complex with other proteins such as SNAP-25 and VAMP enabling the docking and fusion of synaptic vesicles with the presynaptic membrane. This action results in the release of neurotransmitters into the synaptic cleft which then bind to receptors on the postsynaptic neuron to propagate neural signals. Syntaxin 1a ensures efficient synaptic activity and is indispensable for maintaining synaptic plasticity which is critical for learning and memory.
Pathways
Syntaxin 1a is pivotal in the regulation of neurotransmitter release within the synaptic vesicle cycle pathway. This pathway includes the assembly and disassembly of the SNARE complex which are processes vital for neurotransmitter exocytosis. Syntaxin 1a engages closely with proteins like synaptotagmin acting as calcium sensors and Munc18 which helps in initiating SNARE assembly. These interactions highlight the careful orchestration of vesicle trafficking and fusion events necessary for effective synaptic communication.
