Teriparatide (PTH -34) ELISA Kit is a sandwich ELISA for the qunatitative dtermination of Teriparatide (PTH -34) in human plasma and cell culture supernatant.

How the assay works

This assay utilizes a quantitative sandwich enzyme immunoassay format. Streptavidin is pre-coated onto the microwells, where samples and standards are added along with biotinylated antibodies specific to PTH-34, forming an immune complex. A horseradish peroxidase (HRP)-conjugated anti-PTH-34 antibody is then introduced and incubated simultaneously with the samples. Following incubation, the wells are washed to remove any non-specific binding. A TMB substrate solution is added, producing a color change proportional to the concentration of PTH-34 in the sample. The reaction is terminated by adding a stop solution, and absorbance is measured at 450 nm.

Teriparatide (PTH -34) ELISA Kit (ab325032) protocol summary

1. Add 150µL of standard, controls or samples to appropriate wells
2. Add 50µL of Working Antibody solution to each well. Incubate for 180 min at room temperature while shaking at 180-220 rpm
3. Wash
4. Add 200µL of TMB substrate to each well. Incubate for 30 min at room temperature in the dark while shaking at 180-220 rpm
5. Add 50µL of stop solution
6. Read at 450 nm within 10 min
