Notes

Tetracycline is an antibiotic used to treat a number of bacterial infections. It is commonly used to treat acne and rosacea. Historically it was important in reducing the number of deaths from cholera. A broad-spectrum antibiotic of the polyketide class, it is produced by the actinobacterial genus Streptomyces. It acts by inhibiting protein synthesis. It is first-line therapy for rocky mountain spotted fever, Lyme disease, Q fever, psittacosis and lymphogranuloma venereum, mycoplasma pneumoniae and to eradicate nasal carriage of meningococci.

Activity summary

Tetracycline is a broad-spectrum antibiotic that disrupts bacterial protein synthesis. It inhibits the binding of aminoacyl-tRNA to the mRNA-ribosome complex effectively blocking the elongation phase of protein translation. Tetracycline also known as TC is a small molecule with a mass of approximately 444.43 g/mol. It is not naturally expressed in humans as it is a synthetic compound originally derived from Streptomyces bacteria.

Biological function summary

The action of tetracycline affects prokaryotic cells by targeting the 30S ribosomal subunit a component of the bacterial ribosome. Tetracycline prevents amino acids from being added to the growing peptide chain leading to inhibition of protein synthesis. It does not form a complex with other proteins in the bacterial system instead directly interacting with the ribosomal subunit to exert its effects.

Pathways

Tetracycline impacts the translation process a critical component of the genetic expression pathway in bacteria. By blocking the translation pathway tetracycline stalls bacterial growth and multiplication. This effect occurs independently of eukaryotic translation pathways where proteins such as elongation factors EF-Tu and EF-G are involved as tetracycline specifically targets only the bacterial machinery.

Associated diseases and disorders

Tetracycline is relevant in the treatment of bacterial infections such as acne vulgaris and Lyme disease. In acne vulgaris tetracycline helps reduce the bacterial population of Propionibacterium acnes decreasing inflammation and skin lesions. For Lyme disease its effectiveness relates to inhibiting the Borrelia burgdorferi bacteria. Tetracycline does not exhibit direct interactions with proteins associated with these conditions in humans making its actions specific to the bacterial agents causing the disease.
