Required for both basal and inducible autophagy. Determines the localization of the autophagy-specific PI3-kinase complex PI3KC3-C1 (PubMed:18843052, PubMed:19050071). Plays a role in autophagosome formation and MAP1LC3/LC3 conjugation to phosphatidylethanolamine (PubMed:19270696, PubMed:20713597). Promotes BECN1 translocation from the trans-Golgi network to autophagosomes (PubMed:20713597). Enhances PIK3C3 activity in a BECN1-dependent manner. Essential for the autophagy-dependent phosphorylation of BECN1 (PubMed:23878393). Stimulates the phosphorylation of BECN1, but suppresses the phosphorylation PIK3C3 by AMPK (PubMed:23878393). Binds to STX17-SNAP29 binary t-SNARE complex on autophagosomes and primes it for VAMP8 interaction to promote autophagosome-endolysosome fusion (PubMed:25686604, PubMed:37632749). Sandwich ELISA of ab322647 with the capture antibody Anti-ATG14L antibody [EPR28551-44] - BSA and Azide free (Capture) ab322648 dilution at 2 ug/mL and detector antibody Anti-ATG14L antibody [EPR28551-51] - BSA and Azide free (Detector) ab322649 dilution at 0.5 ug/mL.
Interpolated concentrations of native ATG14L in human control wild type HeLa (human cervical adenocarcinoma epithelial cell) and ATG14 (ATG14L) knockout HeLa cell based on 250 ug/mL extract loads. The concentrations of ATG14L were measured in duplicate and interpolated from the ATG14L standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ATG14L concentration was determined to be 397.4 pg/mL in wild type HeLa extract (Human wild-type HeLa cell line Human wild-type HeLa cell line ab255448) and 50.4 pg/mL in ATG14 (ATG14L) knockout HeLa extract (Human ATG14 (ATG14L) knockout HeLa cell line Human ATG14 (ATG14L) knockout HeLa cell line ab264684).
Interpolated concentration of native ATG14L was measured at different sample concentrations. Undiluted samples are as follows: HeLa cell extract (250 ug/mL) and HCT116 cell extract (400 ug/mL). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in antibody diluent CP2 (Antibody Diluent CP2 ab319981). Data represented here are indicative of assay working using example of biological sample. However, we do not guarantee same sensitivity level due to biological variation.
