Function

C3 plays a central role in the activation of the complement system. Its processing by C3 convertase is the central reaction in both classical and alternative complement pathways. After activation C3b can bind covalently, via its reactive thioester, to cell surface carbohydrates or immune aggregates. Derived from proteolytic degradation of complement C3, C3a anaphylatoxin is a mediator of local inflammatory process. In chronic inflammation, acts as a chemoattractant for neutrophils (By similarity). It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes. C3-beta-c. Acts as a chemoattractant for neutrophils in chronic inflammation. Acylation stimulating protein. Adipogenic hormone that stimulates triglyceride (TG) synthesis and glucose transport in adipocytes, regulating fat storage and playing a role in postprandial TG clearance. Appears to stimulate TG synthesis via activation of the PLC, MAPK and AKT signaling pathways. Ligand for C5AR2. Activity summary

C3a anaphylatoxin also known simply as C3a functions as a small protein fragment within the complement system. It forms during the cleavage of complement component 3 (C3) and has a molecular mass of approximately 9 kDa. C3a is mainly expressed on leukocytes endothelial cells and smooth muscle cells. It acts as a potent mediator of inflammation by binding to the C3a receptor (C3aR) found on the surface of various immune cells like mast cells and basophils.

Biological function summary

C3a interacts extensively with cells involved in immune responses. It serves as an inflammatory mediator promoting chemotaxis mast cell degranulation and increased vascular permeability. C3a is not part of a large complex but acts independently to drive immune responses through its receptors. It also plays a role in the process of immune regulation and tissue repair by attracting immune cells to sites of infection or damage.

Pathways

C3a participates in both the complement and coagulation cascades. In the complement pathway it functions alongside other components like C5a and C5b-9 enhancing the local immune responses and pathogen clearance.
