C3a anaphylatoxin also known simply as C3a functions as a small protein fragment within the complement system. It forms during the cleavage of complement component 3 (C3) and has a molecular mass of approximately 9 kDa. C3a is mainly expressed on leukocytes endothelial cells and smooth muscle cells. It acts as a potent mediator of inflammation by binding to the C3a receptor (C3aR) found on the surface of various immune cells like mast cells and basophils.
Biological function summary

C3a interacts extensively with cells involved in immune responses. It serves as an inflammatory mediator promoting chemotaxis mast cell degranulation and increased vascular permeability. C3a is not part of a large complex but acts independently to drive immune responses through its receptors. It also plays a role in the process of immune regulation and tissue repair by attracting immune cells to sites of infection or damage.

Pathways

C3a participates in both the complement and coagulation cascades. In the complement pathway it functions alongside other components like C5a and C5b-9 enhancing the local immune responses and pathogen clearance. The coagulation pathway also involves interactions with various other proteins affecting cell signaling and inflammation bridging innate and adaptive immunity.

C3a contributes to conditions like asthma and autoimmune disorders. In asthma it promotes airway inflammation and hyperresponsiveness by acting on immune cells and tissues. In systemic lupus erythematosus (SLE) C3a levels correlate with inflammation and tissue damage involving proteins in immune complex deposition. Understanding C3a's activity helps in developing therapeutic strategies targeting inflammatory diseases.

Complement C3b binds covalently via its reactive thioester, to cell surface carbohydrates or immune aggregates (PubMed : 6903192).
