Ubiquitin+1 (UBB+1) is an altered form of the ubiquitin protein which plays a significant role in protein degradation. This mutant ubiquitin results from a molecular error during transcription known as a frameshift mutation. The mass of UBB+1 is slightly greater than that of the normal ubiquitin due to an extra amino acid. UBB+1 is expressed primarily in neuronal cells where it accumulates due to its impaired function in tagging proteins for degradation by the proteasome.
Biological function summary

The alteration in UBB+1 affects the cellular protein quality control system. UBB+1 cannot be recognized properly for degradation itself and hinders the ubiquitin-proteasome system (UPS) by competing with normal ubiquitin. UBB+1 does not just influence single proteins; it might also intervene in larger complexes related to proteostasis. Its aberrant accumulation disrupts the clearance of damaged or misfolded proteins which can impact cellular health.

Pathways

Ubiquitin+1 affects several fundamental cellular processes by disturbing the UPS pathway. UBB+1 interacts with proteins involved in the ubiquitin-proteasome pathway such as E1 activating enzymes E2 conjugating enzymes and E3 ligases. Through these associations UBB+1 can induce stress in protein turnover and impair cellular regulation with direct implications in the processes related to protein homeostasis. The effect on these pathways can lead to wider disruptions in cellular signaling and maintenance.

Ubiquitin+1 has a notable impact on neurodegenerative diseases particularly Alzheimer's disease. It is associated with the accumulation of neurofibrillary tangles and plaques seen in these conditions. UBB+1 also connects with proteins like amyloid-beta which play central roles in Alzheimer's pathology. Furthermore UBB+1 accumulation has implications in Huntington’s disease where it links with the aggregation-prone mutant huntingtin protein. These associations highlight the significance of UBB+1 in disease mechanisms involving protein aggregation and cellular stress responses.

Ubiquitin. Exists either covalently attached to another protein, or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of the ubiquitin (polyubiquitin chains) or a linear polymer linked via the initiator Met of the ubiquitin (linear polyubiquitin chains). Polyubiquitin chains, when attached to a target protein, have different functions depending on the Lys residue of the ubiquitin that is linked : Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in proteotoxic stress response and cell cycle; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, DNA-damage responses as well as in signaling processes leading to activation of the transcription factor NF-kappa-B. Linear polymer chains formed via attachment by the initiator Met lead to cell signaling. Ubiquitin is usually conjugated to Lys residues of target proteins, however, in rare cases, conjugation to Cys or Ser residues has been observed. When polyubiquitin is free (unanchored-polyubiquitin), it also has distinct roles, such as in activation of protein kinases, and in signaling.
