Flow Cytometry - Anti-14-3-3 beta antibody [6F6] (AB314175)
Overlay Peak curve showing HeLa (Human epithelial cell line from cervix adenocarcinoma) cells stained with ab314175 (pink line) at 1 : 100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1x10E6cells) for 45min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1 : 200 dilution for 35min at 4°C. Control antibody (green line) was rabbit IgG (1ug/1x10E6cells) used under the same conditions. Acquisition of >10,000 events was performed.

Immunofluorescence staining of PC-3 (Human prostate adenocarcinoma cell line) cells with ab314175 at 1 : 50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor® 494-conjugated Goat Anti-Rabbit IgG(H+L).

14-3-3 beta plays a central role in cellular processes like signal transduction apoptosis and cell cycle control. It often acts in concert with other proteins forming complexes that regulate these vital functions. For instance it binds to Raf-1 kinase affecting cell growth and differentiation. It also interacts with proteins involved in the mitogen-activated protein kinase (MAPK) pathway highlighting its diverse biological significance.
Pathways
14-3-3 beta is important in both the MAPK and apoptotic pathways. In the MAPK pathway it helps regulate the phosphorylation status of proteins involved in cell proliferation. The protein binds to Raf-1 an important component in MAPK signaling impacting cellular responses to growth signals. In the context of apoptosis 14-3-3 beta partners with BAD protein a pro-apoptotic factor sequestering it away from mitochondria this interaction is critical for cell survival by preventing the apoptotic process.
