14-3-3 theta also known as YWHAQ is a member of the 14-3-3 protein family with a mass of approximately 28 kDa. These proteins are known for binding phosphoserine-containing proteins. In the case of 14-3-3 theta it is involved in the regulation of a broad range of proteins by altering activity cellular localization and stability. This protein is widely expressed in many tissues but has high expression levels in the brain indicating its involvement in neural functions.
Biological function summary

14-3-3 theta plays key roles in various signaling pathways and cellular processes. It participates as a scaffold or adapter molecule in the formation of protein complexes aiding in signal transduction. Its ability to bind to numerous client proteins allows it to regulate biological processes such as apoptosis cell cycle progression and cellular stress responses. By facilitating these processes 14-3-3 theta maintains cellular homeostasis and proper cellular responses to environmental cues.

Pathways

14-3-3 theta integrally functions within the MAPK and PI3K/Akt signaling pathways. These pathways involve the regulation of cell growth survival and metabolism. In the MAPK pathway 14-3-3 theta collaborates with proteins like Raf and MEK assisting in the transduction of growth signals. In the PI3K/Akt pathway it interacts with Akt influencing cell survival and proliferation by modulating downstream targets.

Dysregulation of 14-3-3 theta has associations with neurodegenerative disorders like Alzheimer's disease and psychiatric disorders such as schizophrenia. In Alzheimer's disease interactions with tau protein are noteworthy which may affect tau phosphorylation and subsequent aggregation. In schizophrenia alterations in signaling pathways where 14-3-3 theta is an active participant might cause disruptions in neural signaling contributing to the disorder's pathophysiology.
