Function

Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways (PubMed:14578935, PubMed:15071501, PubMed:15644438, PubMed:16376338, PubMed:16959763, PubMed:31024343, PubMed:9360956). Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif (PubMed:35662396). Binding generally results in the modulation of the activity of the binding partner (PubMed:35662396). Promotes cytosolic retention and inactivation of TFEB transcription factor by binding to phosphorylated TFEB (PubMed:35662396). Induces ARHGEF7 activity on RAC1 as well as lamellipodia and membrane ruffle formation (PubMed:16959763). In neurons, regulates spine maturation through the modulation of ARHGEF7 activity (By similarity).

Notes

ab252603 is a BSA and Azide Free antibody supplied in an unconjugated format and it is suitable for sandwich ELISAs to quantify Human 14-3-3 zeta (YWHAZ). The recommended pair for sandwich ELISA is:
Capture: ab252603, Human 14-3-3 zeta (YWHAZ) Capture Antibody (unconjugated)
Detector: Anti-14-3-3 zeta antibody [EPR20639-75] - BSA and Azide free (Detector) ab252662, Human 14-3-3 zeta (YWHAZ) Detector Antibody (unconjugated)
The reference range value is 250 - 8000 pg/mL.

Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. The carrier-free buffer and high concentration allow for increased conjugation efficiency.

This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

The recommended antibody orientation is based on internal optimization for ELISA-based assays. Antibody orientation is assay dependent and needs to be optimized for each assay type. Please note that the range provided for this antibody is only an estimation based on the performance of the product using the recommended antibody pair. Performance of the antibody pair will depend on the specific characteristics of your assay. We guarantee the product works in sandwich ELISA, but we do not guarantee the sensitivity or dynamic range of the antibody in your assay.

Activity summary

The 14-3-3 zeta protein also known as 14-3-3 protein zeta/delta and 1433 protein belongs to the 14-3-3 protein family. These proteins have a molecular weight of around 28-33 kDa. 14-3-3 zeta proteins are expressed across various human tissues including the brain heart and muscles. Mechanically the 14-3-3 zeta protein interacts with different phosphorylated serine and threonine residues on its target proteins. This binding modulates the target proteins' conformation and activity influencing their function in cellular signaling and regulation.

Biological function summary

The 14-3-3 zeta protein plays an important role in several cellular processes. It acts as an adapter in signal transduction pathways and cell cycle control. Additionally the 14-3-3 zeta protein forms complexes with other proteins to regulate apoptosis and stress responses. By binding to various partners this protein influences the distribution and localization of proteins within the cell affecting diverse functions such as metabolism and cell growth.

Pathways

The 14-3-3 zeta protein participates actively in two primary pathways: the MAPK/ERK and PI3K/AKT pathways. Within the MAPK/ERK pathway it influences cellular differentiation and proliferation interacting closely with proteins like Raf and MEK. In the PI3K/AKT pathway 14-3-3 zeta affects cell survival working alongside proteins such as AKT and Bad. These interactions make 14-3-3 zeta a significant player in the regulation of cellular processes.

Associated diseases and disorders

14-3-3 zeta protein has been connected with neurodegenerative diseases and cancer. Altered expression or function of this protein has been observed in conditions like Alzheimer's disease where it interacts with tau protein and in various cancers where 14-3-3 zeta impacts tumor progression by modulating the function of proteins like p53 and Rb. Understanding these interactions helps in exploring therapeutic strategies for disease intervention.
