This ELISA data used the same anti-17-beta-Estradiol antibody clone [EPPTX-R2] in a different buffer formulation (cat# ab215528).

Goat anti-rabbit IgG was coated onto a 96-well plate. Serial dilution of 17β-Estradiol (0, 1, 2, 4, 8, 16, 32 pg/ml) was added (75 μl). Then 3-5 ng/ml of ab215528 (25 μl) was added followed by incubation by shaking for 60 minutes at room temperature. Then 25 μl of HRP-conjugated 17β-Estradiol was added and incubated for 60 minutes at room temperature. After washing, 125 μl Tetramethylbenzidine (TMB) substrate was added and incubated for 30 minutes at room temperature for color development. Then 125 ul of stopping solution (acid solution) was dispensed and OD was read at 450 nm on a microplate reader within 30 minutes.

17-beta-Estradiol also known simply as estradiol or beta-estradiol is a steroid hormone and a member of the estrogen family. It has a molecular mass of approximately 272.39 g/mol. This hormone is synthesized mainly in the ovaries and to a lesser extent in the adrenal glands and the male testes. In cellular mechanics 17-beta-Estradiol functions by entering cells and binding to specific nuclear estrogen receptors alpha (ERα) and beta (ERβ). This binding triggers changes in the cell influencing gene expression and cellular activity.
Biological function summary

17-beta-Estradiol exerts significant roles in reproductive systems such as regulating the menstrual cycle and maintaining pregnancy. It is part of the steroid hormone pathway working closely with proteins like progesterone receptors. Besides its reproductive functions 17-beta-Estradiol impacts bone density cardiovascular health and brain function highlighting its systemic importance. It does not form part of a protein complex but interacts with multiple transcription factors to execute its functions.
