The 2N Tau protein also known as tau or tau protein is a microtubule-associated protein with a mass of approximately 78 kDa. It stabilizes microtubules and is critical for maintaining the axonal transport in neurons. It is highly expressed in the central nervous system particularly in the neurons and becomes hyperphosphorylated under certain conditions. Tau belongs to the family of microtubule-associated proteins and has six isoforms generated through alternative splicing.
Biological function summary

The microtubule-associated protein tau plays a significant role in neuron stability and function by promoting the assembly and stabilization of microtubules. It achieves this through its binding affinity for tubulin which is necessary for the proper functioning of the neuronal cytoskeleton. Tau does not function as part of a complex but its interaction with microtubules is critical for cellular operations including intracellular transport and signaling.

Tau functions in processes essential for cellular transport and microtubule dynamics regulation. It is involved in the MAPK/ERK pathway impacting cellular signaling and stress responses. Tau's regulatory role in microtubule stability connects it with proteins such as tubulin and other microtubule-associated proteins. This interaction ensures proper transmission of signals within neurons and affects cell cycle regulation.

Tau dysregulation and hyperphosphorylation relate closely to Alzheimer's disease leading to neurofibrillary tangles that disrupt neuron function. Additionally tau is implicated in frontotemporal dementia where its misfolding and aggregation significantly impact neuronal survival and function disrupting communication with proteins important for brain function.

Target data

Promotes microtubule assembly and stability, and might be involved in the establishment and maintenance of neuronal polarity (PubMed : 21985311). The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau functions as a linker protein between both (PubMed : 21985311, PubMed : 32961270). Axonal polarity is predetermined by TAU/MAPT localization (in the neuronal cell) in the domain of the cell body defined by the centrosome. The short isoforms allow plasticity of the cytoskeleton whereas the longer isoforms may preferentially play a role in its stabilization.
