Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-5HT3A receptor/5ht3 antibody (AB13897)
IHC image of ab13897 staining in human normal cerebral cortex formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab13897, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
The 5-HT3A receptor acts as a critical mediator of synaptic signalling and neurotransmitter release offering a path for serotonin's effects. As part of a pentameric complex 5-HT3A subunits combine to form functional ion channels. The receptor particularly influences excitatory neurotransmission affecting processes involved in mood regulation nausea and vomiting. Consequently this receptor serves key physiological functions through its modulation of ionic flux and neuronal activity.

The 5-HT3A receptor plays an essential role in pathways related to neurotransmission and neuroplasticity. Within serotonin signalling pathways 5HT3A closely associates with proteins such as serotonin transporter (SERT) elaborating on its role in serotonin uptake and homeostasis. Furthermore its participation in these pathways links to the regulation of calcium-dependent processes interacting with calmodulins and other calcium-binding proteins to influence downstream signalling events essential for cognitive processes and sensory signalling.
