AADACL1 also known as Neuropathy Target Esterase is an enzyme involved in the hydrolysis of cholesterol esters and glycerolipids. It has a molecular mass of approximately 45 kDa. This enzyme localizes mainly to the cytoplasm and expresses highly in the human liver kidney and pancreas with moderate expression in brain tissues. Its role in lipid metabolism suggests importance in maintaining cellular lipid balance.
This enzyme facilitates the breakdown of certain lipid molecules making it integral in lipid homeostasis. As part of a protein complex AADACL1 interacts with other enzymes that regulate lipid metabolism playing a significant role in ensuring cellular lipid composition remains regulated. It helps in preventing the toxic accumulation of lipid substrates in cells.
The enzyme takes part in the neutral lipid metabolism pathway and the lysosomal storage pathway. In these pathways AADACL1 works alongside proteins like lipase and acid ceramidase which contribute to lipid degradation and turnover. Its function ensures lipid molecules get processed efficiently to maintain cellular health.
This enzyme associates with neurodegenerative diseases and metabolic disorders. Mutations or malfunctions in AADACL1 relate to hereditary neuropathy and lysosomal storage diseases. Related proteins such as glucocerebrosidase and sphingomyelinase often interplay with AADACL1 within the context of these diseases linking their functions to disease progression.
